• A5 kb plasmid DNA has two Bam HI sites. You would like to use this
enzyme to digest 5ug of this DNA. How would you do? (DNA
concentration: 2 mg/ml)
Bam HI concentration: 20000 U/ml
Unit definition of Bam HI: 1 unit enzyme can completely digest 1 ug of
lambda DNA at 37 °C in 1 hour. (lambda DNA has 48500 bp and 5 Bam HI
sites)
The reaction needs supplement with 100 ug/ml bovine serum albumin (BSA,
10x concentration)
• Calculate the volume of all ingredients and setup the reaction table.
